Mamelodi Sundowns opened the scoring thanks to a 21st-minute goal from new signing, Brazilian midfielder Lucas Costa only for Ashley du Preez to look to have saved the Glamour Boys from defeat with an 84th-minute equaliser.
However, Masandawana were not to be denied and Neo Maema grabbed a 90th-minute winner.
Kaizer Chiefs now have just one point from their opening two 2023/24 DStv Premiership matches having been held to a goalless draw by minnows Chippa United in their opener last weekend.
By contrast, with six points after two matches, defending champions Mamelodi Sundowns have made a perfect start to the defence of their title having also beaten Sekhukhune United last Friday.
Amakhosi will next be in MTN8 quarter-final action away from home against Cape Town City FC on Sunday, 13 August while Mamelodi Sundowns will take on Swallows in the same tournament on Saturday, 12 August.
